10mM dNTP Mix

  • Effect of dNTP Quality on qPCR: Low Copy Precision IL1-b was assayed by TaqMan® qPCR in 12 replicate reactions containing 25 pg of human genomic DNA (approximately 15 copies) using either Quanta's qPCR-Qualified dNTP Mix or PCR-Grade dNTP mix from a leading supplier. Delayed Cts with higher standard deviation and higher frequency of failed qPCRs was evident in dNTP preparations that did not meet Quanta's rigorous qPCR standards.
  • Effect of dNTP Quality on qPCR TaqMan qPCR of the single-copy gene, IL1-b from log-fold serial dilutions of human genomic DNA (1 mg to 1 pg; ~ 3 x 105 copies to 0.3 copies) using Quanta’s qPCR-qualified dNTP mix (Panel A) or leading supplier’s PCR-Grade dNTP mix (Panel B). Impurities in dNTPs that do not affect classical PCR performance can have detrimental effects on real-time qPCR.

Description

The 10 mM dNTP Mix is a solution of high purity deoxyribonucleoside triphosphates that has been functionally qualified for real-time quantitative PCR (qPCR). It is suitable for use in conventional end-point PCR, real-time qPCR, first-strand cDNA synthesis, as well as other applications that require dNTP as substrate. We have found that dNTP quality and purity can directly affect dynamic range, sensitivity and precision of qPCR assays. Surprisingly, dNTP preparations that fail our qPCR functional testing are often >99% triphosphate purity by HPLC analysis and function perfectly well in conventional end-point PCR assays, including amplification of long (up 20 kb) templates. The use of qPCR certified dNTPs is a simple but important measure to ensure consistent assay performance.

Storage conditions

Store the 10 mM dNTP mix at -20°C in a constant temperature freezer. Avoid repeated freeze/thaw cycles. When stored under these conditions the dNTP mix is stable for one year.
